reports no longer work

After no changes except custom fields in some modules suddenly reports (5.0.0f Pro) can no longer be modified or created. Symptoms:
-Add related button does not work/show nothing
- Add new filter button does nothing
- add column button does nothing
- in choose display columns no columns are displayed
- in already created reports the report is shown, but can not be modified with the same symptoms.
If I open reports with IE i get an script error in IE "Error: module_defs is undefined, if this is of any help. Log file does not show anything I have also tried repair function, which does not change anything Please help!

We had the same error on our hosted environment. Check the apache error log. We had a permissions issue with the one of the .js files in the cache/modules directory.
