Second mailer to campaign

Hello,
Last week I created an email campaign and sent this out, I had some response, but nothing great. What is the procedure of doing a second email campaign to the same target, ensuring that the OptOuts are not included?

Do I have to remove the OptOuts from the Target list manu?

Can I include another target list to the second campaign?

Please tell me if I am misunderstanding this,

I would like to create a followup email campaign, but I am not sure if I should use the same Campaign or create a new one.
